Quantity:5 ml

Working concentration:neat

Presentation:Antibody solution in stabilizing phosphate buffer pH 7.3. Contains 0.09 % sodium azide**. The volume is sufficient for at least 50 immunohistochemical tests (100 µl working solution / test). Use appropriate antibody diluent e.g. BIOLOGO Art. No. PU002, if further dilution is required.

Immunogen:Cytokeratins isolated from human mamma carcinoma cell line MCF-7

Specificity:Human intermediar filament cytokeratins 8 and 18 (MW 52,5 resp. 45 kDa)

Species Specificity:Human, cat, lizard, sheep

General Information:Cytokeratins are a group of water-insoluble filamentous proteins. They are constituents of the cytoskeleton of epidermal cells and other epithelial cells. Up to now 20 different cytokeratins have been characterised by gel electrophoresis. They have been subdivided into a basic and an acid subfamily. Most frequently the nomenclature according to Moll et al. (1982) is used. CK 8 is an acid CK, which is expressed by most epithelia except squamous epithelia. It is frequently expressed together with CK 18 (1,2). Cytokeratin 18 is expressed by simple epithelia of the intestinal tract, lung, bronchi, urinary tract, liver and gland ducts.

Special properties:Clone 5D3 labels low molecular weight cytokeratins 8 and 18 in all simple epithelia, including glandular epithelia of the thyroid, breast, skin and epithelia of the gastrointestinal tract. It doesn't stain squamous epithelia and is used for the differentiation of carcinoma versus lymphoma. It will also detect micro-metastases in bone marrow.

Positive control:Skin (sweat glands)

Pre-treatment:In formaldehyde-fixed material pre-treatment with trypsin (0,1% in TBS containing 0,1% CaCl2, pH 7.8) 15 -30 min at 37°C is recommended.

Applications:C, P

Incubation time:60 min at RT

Secondary reagents:We recommend the use of BIOLOGO's Universal Staining System DAB (Art. No. DA005) or AEC (Art.-No. AE005), if higher sensitivity is required VECTASTAIN Elite ABC (Art. No. PK-6102) or ABC AP (Art. No. AK-5002) systems are applicable.

Storage:2-8°C

Notes:
*BIOLOGO's antibodies are intended for in vitro research use only. They must not be used for clinical diagnostics and not for in vivo experiments in humans or animals.

** The preservative sodium azide is known to be poisonous and potentially hazardous to health. It should be handled only by trained staff. Despite of the product's low azide concentration it must be handled with care. Dispose according to regional rules!
